Application des techniques moléculaires de PCR-RFLP et de PCR-RFLP-SSCP dans l'étude de la différenciation génétique de deux huîtres creuses : Crassostrea gigas et Crassostrea angula ArchiMer

Les 2 taxons d'huîtres creuses C. angulata et C. gigas n'étant pas différenciables sur des critères morphologiques, écologiques ou physiologiques, l'étude de leur passé évolutif fait appel à l'utilisation de marqueurs génétiques. L'étude du polymprohisme des séquences mitochondriales codant pour la sous-unité I de la cytochrome oxydase par la technique de PCR-RFLP a été réalisée sur 197 individus appartenant à 5 populations d'origine géographique différente, de type Crassostrea angulata ou Crassostrea gigas. As principais espécies de vírus envolvidas na etiologia do enrolamento da folha da videira (Vitis spp.) são Grapevine leafroll-associated virus 1 e 3 (GLRaV-1 e -3). Neste estudo da variabilidade desses vírus, foram amplificados dois fragmentos de DNA (396 bp do GLRaV-1 e 602 bp do GLRaV-3) por RT-PCR, a partir de RNA total extraído de nervuras e pecíolos de videiras infetadas, utilizando-se dois pares de oligonucleotídeos. Bacterial ecology of abattoir wastewater treated by an anaerobic digestor BJM

Abstract Wastewater from an anaerobic treatment plant at a slaughterhouse was analysed to determine the bacterial biodiversity present. Molecular analysis of the anaerobic sludge obtained from the treatment plant showed significant diversity, as 27 different phyla were identified. Firmicutes, Proteobacteria, Bacteroidetes, Thermotogae, Euryarchaeota (methanogens), and msbl6 (candidate division) were the dominant phyla of the anaerobic treatment plant and represented 21.7%, 18.5%, 11.5%, 9.4%, 8.9%, and 8.8% of the total bacteria identified, respectively. Isolates of Citrus tristeza virus around the world differ widely in their biological properties. These properties may depend on strain mixtures within individual trees. Monoclonal Antibody 13 (MCA13) is an antibody that detects almost all the severe strains causing Quick Decline and Stem Pitting CTV syndromes. The objectives of this work were: I) to determine the percentage of CTV isolates reactive to the MCA13 antibody in each Colombian region evaluated and II) to analyze the genetic diversity of isolates from these regions. El maíz (Zea mays L.) de calidad proteínica (QPM) es nutricionalmente superior en comparación con el maíz normal, porque produce semillas con casi el doble de los aminoácidos (aa) lisina (Lys) y triptófano (Trp), esenciales para la síntesis endógena de la vitamina B niacina. Detection of point mutations by non-isotopic single strand conformation polymorphism BJMBR

We have developed a procedure for nonradioactive single strand conformation polymorphism analysis and applied it to the detection of point mutations in the human tumor suppressor gene p53. The protocol does not require any particular facilities or equipment, such as radioactive handling, large gel units for sequencing, or a semiautomated electrophoresis system. This technique consists of amplification of DNA fragments by PCR with specific oligonucleotide primers, denaturation, and electrophoresis on small neutral polyacrylamide gels, followed by silver staining. The sensitivity of this procedure is comparable to other described techniques and the method is easy to perform and applicable to a variety of tissue specimens.

The influence of cytoplasmic inheritance on birth and weaning weight was evaluated in an experimental Hereford herd. Data on 1,720 records for birth and weaning weights from calves born between 1963 and 2002 were studied. Variance components were estimated using MTDFREML procedures and an animal model was fitted for each trait. Direct and maternal additive effects and permanent environment and maternal lineage effects were treated as random, while year and month of birth, age of dam and sex of the calf were treated as fixed. Se desconoce la etiología de la enfermedad muerte súbita de los cítricos (MSC) y su relación con una variante severa del virus tristeza de los cítricos (Citrus tristeza virus, CTV). Para estudiar una posible relación se estudiaron cinco poblaciones de 100 árboles cada una, en regiones con y sin presencia MSC, en huertas comerciales de los estados de São Paulo y Minas Gerais, Brasil. Large-scale mortality events have been observed in Pacific oyster Crassostrea gigas on the west coast of France since the early 1980s, particularly during summer. In order to understand the causes of this mortality, two generations of oysters from single-pair matings were studied in three sites on the French Atlantic coast (Baie-des-Veys, Auray and Ronce-les-Bains). Isolation and sequencing of doublesex/male abnormal 3 (DM) related transcription factor (Dmrt) genes from the Asian toad Bufo gargarizans (Cantor, 1842) Genet. Mol. Biol.

The doublesex/male abnormal 3 (dsx/mab-3 or DM) domain gene family involved in sexual development encodes putative transcription factors including a DNA-binding homology motif, the DM domain. We used highly degenerate primers to clone and sequence seven distinct DM related transcription factor (Dmrt) genes from the Asian toad (Bufo gargarizans Cantor, 1842). Isolation and sequencing of seven Sox genes from the lacertid lizard Eremias breuchleyi Genet. Mol. Biol.

The Sox family of genes shares a high sequence similarity with the HMG box region of the human Y chromosomal gene, SRY. We used highly degenerate primers to clone and sequence seven Eremias breuchleyi Sox genes (EbSox2, EbSox3, EbSox4, EbSox11, EbSox12, EbSox14 and EbSox21). A database search for the cloned sequences revealed the following percentage identity with the homologous human SOX genes: EbSox2 = 96%, EbSox3 = 88%, EbSox4 = 94%, EbSox11 = 99%, EbSox12 = 96%, EbSox14 = 98%, EbSox21 = 97%. Cluster analysis indicates that they seem to belong to group B and group C of Sox gene family, respectively.

Sox (SRY-related HMG-box) genes encode a family of transcriptional regulators, which are characterized by a conserved 79-amino acid domain known as HMG-box. They play essential roles in a diverse range of processes including sex determination and the development of the central nervous system (CNS), neural crest and endoderm. Within this abundance of proteins, kappa-casein is noteworthy, since it has been associated with differences in milk yield, composition and processing. Human papillomavirus (HPV) infections of the high-risk types are strongly linked to the development of cervical carcinoma. The HPV oncoproteins E6 and E7 are thought to play a crucial role in this process through their interactions with the p53 protein and the retinoblastoma susceptibility gene product pRb, respectively. E6 binds to p53 protein promoting its degradation. This is considered to contribute to the oncogenesis of HPV-associated anogenital cancer. Mutation and association analysis of the PVR and PVRL2 genes in patients with non-syndromic cleft lip and palate Genet. Mol. Biol.

Orofacial clefts (OFC; MIM 119530) are among the most common major birth defects. Here, we carried out mutation screening of the PVR and PVRL2 genes, which are both located at an OFC linkage region at 19q13 (OFC3) and are closely related to PVRL1, which has been associated with both syndromic and non-syndromic cleft lip and palate (nsCLP). We screened a total of 73 nsCLP patients and 105 non-cleft controls from the USA for variants in PVR and PVRL2, including all exons and encompassing all isoforms. We identified four variants in PVR and five in PVRL2. One non-synonymous PVR variant, A67T, was more frequent among nsCLP patients than among normal controls, but this difference did not achieve statistical significance.

The promoter region and the beginning of the coding region of the hsp70 stress gene were analysed in broiler chickens of a commercial breed (Hubbard-Pettersen), a breed selected for weight gain (PP1) and a non-selected breed (naked-neck Label Rouge).
